Séquençage et annotation de génomes : les membres du laboratoire BF2I ont été impliqués dans plusieurs projets de séquençage. Ils ont participé au séquençage et à l’annotation du génome du puceron du pois (IAGC, 2010, Wilson et al., 2010, Gerardo et al., 2010), et du génome de la bactérie symbiotique de Sitophilus, Sodalis pierantonius (Oakeson et al., 2014, GBE). Ils sont actuellement en train de coordonner le séquençage du génome de Sitophilus oryzae, en collaboration avec le groupe d’A. Latorre (Instituto Cavanilles de Biodiversidad y Biología Evolutiva Universitat de València, Espagne).

Découverte d’un nouveau peptide antimicrobien (ColA) qui est impliqué dans le contrôle du symbiote chez le charançon. Le peptide ColA cible spécifiquement les symbiotes dans les cellules bacteriocytaires, inhibe leur division cellulaire et leur croissance et limite leur espace au bactériome (Login et al., 2011, Science).

Identification des gènes et des voies clés du métabolisme de la symbiose puceron/Buchnera : En combinant des approches métaboliques et transcriptomiques, nous avons identifié une voie clé du métabolisme symbiotique : la voie de la tyrosine (Rabatel et al., 2013). Partagée entre le puceron du pois et son symbiote primaire B. aphidicola, elle régule le développement parthénogénetique de l’insecte. Dans cette voie, nous avons mis en évidence la fonction centrale de la phénylalanine hydroxylase, dont l’inactivation génique affecte sévèrement la fécondité des pucerons et le développement embryonnaire (Simonet et al., 2016, Scientific Reports). 

Découverte du nouveau mode d’action d’un bioinsecticide PA1b : nous avons montré que le peptide PA1b cible et bloque la pompe à proton vacuolaire (V type H+-ATPase). Comme l’activité de la V-ATPase est essentielle pour l’absorption des nutriments énergétiques dans l’intestin de l’insecte, l’utilisation de cette nouvelle propriété de PA1b est un moyen prometteur pour lutter contre les insectes nuisibles (Chouabe et al., 2011).

Découverte d’un nouvel homologue de PA1b à haute activité insecticide : les analyses phylogénétiques de PA1b chez Medicago truncatula ont révélé l’existence d’un nouveau homologue de PA1b, nommé AG41. Ce dernier a montré une activité insecticide très forte et inattendue.
